CRISPR may be the key to freeing mankind from many diseases.
In 2006, was described in the case of relatively rare genetic diseases. One boy, a native of Pakistan, found a very interesting deviation: he could feel warmth, cold, texture items, no problems were detected touch, but did not feel pain. Unfortunately, this boy is no longer alive, because he so believed in his «invulnerability» that in trying to prove it I jumped off the roof. However, the group managed to carry out some research of the genome of a young Pakistani man, and today these data will help to create painkillers that would act at the genetic level. They will work better than the opiates and will not cause addiction.
As you know, in order to develop a similar painkillers, scientists had to use . A new approach to getting rid of the pain mimics a rare mutation of a Pakistani boy. And it has already been tested on mice in the laboratory of Prashant Mali, University of California at San Diego.
The Study, which is for temporary blocking of pain, held in July, but the scientists didn't want to give any comments until then, until all the data of their research will not pass a full background check. The basic idea is to introduce into the cerebrospinal fluid viral particles carrying the modified version of the CRISPR designed to interrupt pain signals. Modifications (or temporary suspension) is subjected to a gene called SCN9A which is responsible for the production of special molecules (it is called Nav1.7). She, in turn, is a key factor in the passage of pain impulse to the brain.
Evidence of the dominant role of this gene and the corresponding molecules were obtained from people with a strange inherited diseases that cause a decrease in pain threshold. Already mentioned in the beginning of the article Pakistani boy was a family member with a mutation that completely disable the gene SCN9A. Thanks to them, managed quite well to study this mutation. But until now, attempts to mimic this mutation did not succeed, but researchers from California have succeeded.
The Data obtained during the experiments on animals show that completely remove the pain syndrome is not yet possible. But to significantly reduce its manifestation have already quite capable. The effect of therapy, CRISPR compare with the use of opiates. Except for the fact that CRISPR is not addictive. This is a huge step forward.
Computer simulation of how CRISPR works with «cutting» DNA fragments
Currently, the researchers plan to begin full-fledged research with the participation of the people. Experts say that such gene therapy can help to relieve chronic pain a great number of people. For example, patients in the later stages of cancer, or people with congenital neuralgia. Also it is impossible to exclude and more «standard» application of a new type of painkillers: the use of CRISPR-medication during surgery or in patients with serious injuries, for example, after accidents or in war zones. What other applications you think you can find such anesthetic? Tell us about it in our
For more quick marketing scientists Navega Therapeutics, which will cooperate with the FDA (Management on sanitary inspection behind quality of foodstuff and medicines ) and a number of research centers in order to check the health of the new drug.
